  "abstract": "Objectives Despite a robust drug development pipeline, lupus remains far behind other rheumatic and autoimmune conditions for which dozens of targeted therapies have been approved. Complex challenges engender this state requiring a bold, collaborative approach such as a Public-Private Partnership (PPP). Lupus Accelerating Breakthroughs Consortium (Lupus ABC), a PPP established in 2023, unites the lupus community to identify and address, in the pre-competitive setting, the most pressing impediments to lupus drug development. Here, we provide an update on the impactful accomplishments of Lupus ABC to date.Methods Currently, Lupus ABC has nearly 50 member groups, including lupus patients and their care partners, representatives from industry, academia, FDA (CDER and CBER), other government and non-profit organizations and professional societies (ACR and CARRA). A robust governance structure, with patients at its center, guides and supports Lupus ABC activities which are implemented through multiple Working Groups (WGs). Initial projects were selected from LABC members, but a recent request for proposals has expanded Lupus ABC’s reach to include the global community.Results Lupus ABC’s effective operationalization (PMID: 40169881) and rapid launch of three WGs and one partner project (TRM-SLE) has already yielded key advances. Most notably, the CLASI WG led to the FDA acknowledgement that CLASI 70 is a reasonable instrument for assessing treatment response in Cutaneous Lupus Erythematosus (CLE) and can be used to establish efficacy in CLE trials (in press). The CAR-T WG developed key considerations for the conduct of clinical trials in this area (PMID: 41052891), while the PRO WG convened an impactful meeting with actionable outcomes (submitted). Last quarter, Lupus ABC launched two new PRO projects and held two exploratory meetings on immune resetting therapies that generated remarkable community engagement and led to the development of a new project on this topic.Conclusions Lupus ABC offers an effective framework for collaboratively addressing common challenges in lupus drug development by identifying shared solutions. In just two and a half years, Lupus ABC has made significant contributions to the field demonstrating its unique value and vast potential.
