  "abstract": "Background CD24 is a heavily glycosylated glycosylphosphatidylinositol (GPI)-anchored protein expressed on the surface of many cell types including cancer cells. It is overexpressed in many cancer types and is known to serve as a ‘don’t eat me’ signal to macrophages through its engagement of the inhibitory receptor Siglec-10. 1 Methods We analyzed CD24 copy number, mRNA and clinical data across all indications in The Cancer Genome Atlas (TCGA). We also analyzed CD24 copy number and mRNA of all cell lines in DepMap.Results We found that CD24 copy number gain is most frequently observed in ovarian cancer, sarcomas, and non-seminomatous germ cell tumors (>30% of tumors showed CD24 gain); followed by endometrial, colorectal, and miscellaneous neuroepithelial cancers (>24%). CD24 copy number correlates to mRNA in many cancer types. CD24 gain is significantly associated with poor overall survival in breast, 2 cholangiocarcinoma, endometrial, esophagogastric, glioma, and renal cancers. CD24 gain is prevalent in the uterine carcinosarcoma subtype of endometrial cancers (59% vs. 24% overall) and basal-like subtype of breast cancers (47% vs. 21% overall). Small cell lung cancer (SCLC) is an aggressive cancer with limited biopsies thus it is absent in TCGA. We analyzed all 1,925 cancer cell lines in DepMap and found that CD24 gain is frequent in SCLC lines over non-small cell lung cancer (NSCLC) or other non-lung cancer cell lines.
