  "abstract": "Background Treatment of triple negative breast cancer (TNBC) is limited to chemotherapy, with immunotherapy showing promising yet partial results. 1 One contributor to an effective response to immunotherapy, is the immunogenicity of cell death caused by chemotherapy. This work aims to determine the platin-based chemotherapies that can produce an immunogenic cell death in a syngeneic orthotopic mouse model.Methods The ”gold standard” assay to evaluate the ability of dying cells to trigger an adaptive immune response is the prophylactic vaccination of immunocompetent syngeneic animals. 2 In this study, we use the syngeneic orthotopic model TNBC to determine if platin or anthracycline chemotherapy treatments generate an immunogenic cell deathResults We reveal platin-based chemotherapies are more effective in generating an immunogenic cell death, with mice that were inoculated with platin treated cells had delayed or no tumor onset when challenged with live cancer cells. Within platin-based chemotherapies, oxaliplatin treatment results in increased ATP release and calreticulin exposure compared to cisplatin.Conclusions We show the syngeneic orthotopic model of TNBC is an effective model to evaluate immunogenic cell death with the vaccine-like assay. In this model, we distinguish oxaliplatin as more immunogenic than other chemotherapies.References Britten K, Bardia A, McAndrew N.
