  "abstract": "Introduction Resistance-based training (RBT) is effective in maintaining muscle function and metabolic health during aging. However, its impact on systemic metabolic remodeling and the potential modulatory effects of polyphenol supplementation (PS) are not well defined. This study aimed to investigate whether RBT combined with PS alters the resting plasma metabolome in older adults.Materials and Methods 41 healthy adults (55–70 years, mean±SD=61±4years) participated in a 12-week supervised RBT program. Participants received either a PS (n=20) or placebo (n=21) for 30 days prior to and throughout training. Resting plasma samples were collected before intervention (Screen), after 30 days of PS (Pre), and following 12 weeks of RBT (Post). Metabolites were extracted using a methanol-based protocol and analysed by hydrophilic interaction liquid chromatography coupled to high-resolution Orbitrap Exploris 120 mass spectrometry (HILIC-LC-MS). Data preprocessing and statistical analyses were performed in MetaboAnalyst6.0.Results No major metabolic differences were observed from Screen to Pre in either group, indicating no preload effect. In the polyphenol group, training (Post vs Pre) led to higher L-cystine, citrulline, ethanolamine, and L-acetylcarnitine (p < 0.05), metabolites linked to mitochondrial substrate utilization, nitric oxide metabolism, and redox regulation. In contrast, placebo group showed increases in L-alanine, L-acetylcarnitine, and butyrylcarnitine.Conclusion Twelve weeks of RBT modulated the resting plasma metabolome in older adults. Both groups showed training-related metabolic remodeling, but the polyphenol group exhibited fewer acylcarnitine accumulations and higher levels of metabolites linked to mitochondrial oxidation and redox regulation, indicating more complete β-oxidation and improved metabolic efficiency.",
