  "abstract": "Purpose The aim of this study is to evaluate the quality of DEMK graft preparation in the International Eye Bank of Prague.Methods Retrospective analysis of DMEK grafts preparation in our eye bank, focused on donor age, death to preparation time, average endothelial cell density prior and post preparation and time needed for preparation. We also analysed percentage of graft preparation failure associated with donor medical history.Results Since March 2018 till September 2024 1116 DMEK grafts were pre-cut in the International Eye Bank of Prague. All preparations were performed by 3 technicians and distributer in to 9 hospitals. The average age of donors used for DMEK preparation was 64 years of age, average death to preparation time was 5 days. Endothelial cell density prior preparation was on average 2902 ± 298 cells/mm2 and 2856 ± 265 cells/mm2 post preparation. Average time needed for preparation of 1 DMEK graft was 7 minutes. Percentage of graft preparation failure was 3,2%. In 49% of unsuccessful DMEK preparation we were able to find diabetes mellitus in donor’s medical history.Conclusions We believe that pre-cut DMEK grafts prepared in our eye bank represent tissues of high quality and safety. Percentage of graft preparation failure is very low and is at least in 49% associated with diabetes mellitus in donor’s medical history.",
