  "abstract": "Purpose Tissue-C (AL.CHI.MI.A. SRL) is a corneal culture medium intended for the storage of human cornea at 31°C for up to 30 days. The device contains 100 mL of medium packaged in borosilicate glass vials with a sterile butyl rubber closure and an aluminum capsule. During corneal culture at 31°C, the medium volume may vary significantly after withdrawing samples for microbiological evaluations. The objective of this study was to investigate the effects of different volumes of Tissue-C during corneal storageMethods Six (n=6) human corneas unsuitable for transplantation were employed for this study and divided in three experimental groups (n=3 tissues each): corneas stored in 100 mL (control) or 80 mL of Tissue-C medium. All corneas were stored at 31°C for 30 days. Corneal quality parameters were evaluated before (Day 0), during (Day 14) and at the end (Day 30) of the storage in Tissue-C and additional examination was performed after transferring the corneas in deswelling medium Carry-C (AL.CHI.MI.A. SRL) for an additional 24h incubation (Day30+1) at room temperature (RT). Endothelial Cell (EC) Density (ECD) was determined by light and specular microscopies techniques. EC mortality and EC morphology score were determined after Trypan Blue staining. EC morphological parameters CV% (coefficient of variation) and HEX% (hexagonality index) were also determined with specular microscopy. EC viability was determined by fluorescent microscopy after Calcein-AM staining. An additional assessment of endothelial layer morphology was performed with light microscopy at the final time point (Day 30+1) after the non-vital staining with Alizarin Red. Optical coherence tomography (OCT) was employed to measure central corneal thickness (CCT, Day 0 and Day 31+1) and corneal transparency (Day 0 and Day 31+1) was evaluated with a lux-meter.Results ECD, EC mortality, EC viability, CV% and HEX% were comparable in the corneas stored in 80 mL and 100 mL volume of Tissue C. No differences in CCT and corneal transparency were shown in among the groups after deswelling phase in Carry-C.Conclusions According to the obtained results, 100 mL and 80 mL of Tissue-C medium can preserve human corneas at 31°C for 30 days.",
